A BRIEF SUMMARY OF M.Sc (AGRICULTURE) THESIS: Title of the Thesis: In vitro propagation and artificial seed production of some orchid species The experiment was carried out in the Tissue culture laboratory of Assam Agricultural University in the year 1996 to 1998. The experiment was carried out by taking the capsules/embryos of six different orchid species namely Cymbidium aloifolium, Cymbidium pendulum, Arundina bambusifolia, Spathoglottis plicata, Rhynchostylis retusa, Acampe longifolia. Hybridization was carried out among different orchid species. But, only these two hybrids viz Dendrobium moschatum x Arundina bambusifolia and Dendrobium infundibulum x Arundina bambusifolia were able to produce viable seeds. The germination of the embryos of these six species and two hybrids and their subsequent regeneration were tested utilizing two different media, namely Nitsch and Knudson C as basal media fortified with vitamin, growth regulators and organic extracts like malt extract, tomato juice and coconut water. Some of the protocorms formed from embryo culture were utilized for full plant regeneration and some protocorms were utilized for preparation of artificial seeds using gel matrix technique. Investigations were also carried out to study the storability and viability of seeds stored in different storage conditions such as at room temperature, at 4oC and at -2oC with or without liquid paraffin. The seedlings regenerated from embryo culture and artificial seed reculture were hardened and successfully transferred to field condition. The study on hardening requirement of tissue-cultured seedlings prior to transferring into main field revealed that some species of orchids could be transferred to main field without hardening.
