MOLECULAR CHARACTERIZATION OF PAPAYA RINGSPOT VIRUS
Highlights of research MSc. work
Molecular characterization of papaya ringspot virus was carried out for PRSV from Parbhani locality (India).
1)Maintains of PRSV Parbhani isolate was done on papaya, cucumber squash and pumpkin and its’ symptoms were studied.
2)Electron microscopic studies reveled to be flexuous rod shaped particles measuring about 750x12 mm in size.
3)Serological studies were carried out using coat protein specific antibodies using ELISA,
4)Molecular weight of coat protein was approximately 31 Kda using
SDS- PAGE.
5)Coat protein gene of size 1.3 Kb was isolated using PRSV specific primers, which were gel elutated and cloned into pTZ57R plasmid and multiplied in DH5α E coli strain for sequencing.
6)Sequencing coat protein reveled 1479 nucleotide long sequence with specificity of DAG motif at 150th position which is important of aphid transmissibility, and showed 83.5 to 98.2% similarity at nucleotide level with other published Indian isolates.

Qualitative and quantitative ELISA analysis of Bt cotton for Cry1Ac, Cry1F gene(Widestrike)
Qualitative ELISA analysis of Bt Brinjal for Vip, Cry2Ab gene.
Qualitative ELISA analysis of Bt Okra for Cry1Ac gene.
3)PCR analysis of transgenic plants.
Analysis of transgenic Cotton for Cry1Ac, trangenic Brinjal for Vip gene.
4)Blotting analysis to find out copy number of inserts in transgenic plants.
Southern Blotting analysis of transgenic Brinjal to find out copy number of Vip gene.
